The effect of gene therapy with nanoliposomes containing plasmid DNA encoding Interleukin-12 in the regression of tumor masses in mouse

Abstract
Purpose: The purpose of this study is to investigate effect of gene therapy with nanoliposome containing DNA Plasmid encoding IL-12 in regression of murine CT-26 colon carcinoma cells. Procedure: pmIL-12 was extracted by utilization of the DNA preparation Kit and then purified. Then, nanoliposomes containing pmIL-12 was prepared with the freeze drying monophase solution technique and thru utilization of DOTP, DOPE, and Cholesterol lipids. After determination of distribution of particles, Zeta potential, and morphology of the prepared liposome, its effect on regression of colon cancerous cells of murine was evaluated. For this, CT-26 cells were injected subcutaneously into murine. When the volumes of tumors were measured to be approximately 90 to 100 mm3, nanoliposomes were injected intra-tumorally. the sizes of tumors were measured and compared to the tumor volumes of the control group. Findings: The size of particles were in the range of 123.4 and 297.9 nm. Based on the obtained results from the TEM, the prepared liposomes were spherical and their size range was in nano. The regression of tumor sizes by utilization of nano-liposomes containing pmIL-12 were greater than the naked DNA. Conclusion: Thru utilization of enhanced freeze-drying of monophase solution, it is possible to produce nanoliposomes containing DNA with high encapsulation efficiency and enhanced anti-tumor effect in comparison to naked DNA. Based on the results, nanoliposomes containing IL-12 which is prepared by freeze-drying of monophase solutions method and with the least cytotoxicity, could reduce tumor size in tumor bearing mice compared to naked DNA.
