Evaluation of effects of Troxerutin on the biochemical factors against Doxorubicin-induced cardiotoxicity in rats

Abstract
Introduction: Doxorubicin (DOX) an anthracycline antibiotic, is one of the most effective anticancer agents which it’s utilization is limited because of cardiotoxicity induction. Troxerutin is a natural bioflavonoid which has antioxidant and antiinflammatory effects. Aim: The aim of this study was to evaluate the protective effect of TRX against doxorubicin-induced cardiotoxicity in rat. Method: In this study, male wistar rats (200-250 g) were used. The animals were randomly devided into 4 groups with 6 animals in each group. Normal control group just received normal saline. DOX-treated group were injected with Dox (2 mg/kg, i.p.) in six equal injections at 48 h intervals and pretreatmented groups with troxerutin which animals received TRX (100 mg/kg , 200 mg/kg i.p.) firstly for one week and then from 8th day rats received TRX and DOX with the same protocol which has described for DOX administration. After 24 h of the last injection, animals were anesthetized with intraperitoneally injection of 100 mg/kg ketamine and 10 mg/kg xylazine. After the surgery cardiac tissue and blood samples were collected. Results: Treatment with troxerutin specially with dose of 200 mg/kg could improve disorders caused by doxorubicin. Biochemical markers such as TAC , SOD, GPx, which decreased during the administration of DOX , increased with troxerutin administration as well as cTnI and MDA cardiac biomarkers, which their serum and tissue level were increased while administration of doxorubicin, decreased with subsequent administration of troxerutin. Conclusion: Troxerutin with anti-inflammatory and antioxidant mechanism can significantly reduce the toxicity of doxorubicin-induced cardiotoxicity by affecting biochemical parameters.
