The Relationship between Apelin-13 and microRNAs related with diabetes, in vitro and obese patients with type 2 diabetes

The purpose of this study was to measure apline-13, and 375 and 107 microRNAs in the serum of patients with newly diagnosed type 2 diabetes mellitus, as well as healthy people. The relationship between sex hormones, peripheral serotonin and biochemical factors with apelin-13 was analyzed. The role of apelin-13 in a laboratory model was also studied.
Methods
In this study, 50 obese women with type 2 diabetes and 50 healthy obese women, as control group were selected. Apelin-13 concentrations, peripheral serotonin, insulin and sex hormones were measured by ELISA technique. The expression levels of miR-375 and miR-107 in serum were evaluated using qPCR technique. Apoptosis was evaluated in the CRI-D2 pancreatic cell line treated with Apelin-13 by flowcytometry. Data were analyzed by version 16 of SPSS software.
Results
Apelin-13 levels (P = 0.012) and the expression levels of miR-375 and miR-107 were high in the serum of obese diabetic women compared to control group. There was no association between apelin-13 and the mentioned microRNAs. The association between serotonin (P < 0.001), estimated average glucose (P = 0.02) and insulin (P = 0.03) with apelin-13 were statistically significant in the diabetic groups. Apoptosis was induced in CRI-D2 cells treated with apelin-13.
