Abstract
IntroductionIt is well known that antioxidant reserve and antioxidant enzyme capacity are significantly reduced after ischemia and reperfusion and this makes the heart susceptible to damage caused by ischemia and reperfusion.ObjectivesIn the present study, effects of hydroalcoholic extract of grape seed (GSE) on ischemia-reperfusion (I/R)- induced infarct size and cardiac arrhythmias were investigated in isolated rat heart.Materials and MethodIsolated rat hearts were divided into four groups (n = 8-12 in each group) and mounted on a Langendorff apparatus at constant pressure, then subjected to 30 min regional ischemia followed by 120 min reperfusion. In control group, the hearts were perfused by a modified Krebs-Henseleit solution throughout the experiment, however, test groups were perfused by enriched Krebs solution with GSE (1, 10 and 100 إg/ml) during I/R. The ECGs were recorded and analyzed to determine cardiac arrhythmias inclusive number of total ventricular ectopic beats (VEBs), number of ventricular tachycardia (VT), duration and incidence of VT and ventricular fibrillation (VF) . The infarct size was determined by triphenyl tetrazolim chloride and computerized planimetry methods. ResultThe results demonstrated that perfusion of GSE produces significant reduction in infarct size in isolated rat heart. In the control group, the infarct size was 315 %, while perfusion of GSE (1, 10, and 100 إg/ml) reduced it to 123 (p<0.05), 166 (p<0.05) and 21 % (p<0.01), respectively. The extract (1 إg/ml) caused significant reduction (p<0.05) in the number, duration and incidence of ischemic VT. At the reperfusion phase, low concentrations of GSE (1 and 10 إg/ml) showed significant reduction (p<0.05) in the number of VEBs and number, duration and incidence of VT .Discussion and conclusionResults of this study showed cardioprotective effects of GSE against I/R injuries by reduction of infarct size and arrhythmias.
