Abstract
In this research, the effects of Danae Racemosa extract (DRE) against oxidative stress in isolated rat hepatocytes has been tested. First, the maximum non-toxic concentration of extract determined by dose response experiments. Then the RC50 value was calculated by DPPH test and compared with Rutine RC50 as an standard. Then using 4 experiments (trypan blue exclusion, ROS formation, mitochondrial membrane potential and the hypoxia) the protective effect of extract towards hepatosytes against toxic substance was evaluated. The DRE solution at the concentration of 200 was found as the maximum non-toxic concentration. The methanol extract of aerial organs in Danae racemosa L. in DPPH Test, showed a significant antioxidant activity (RC50=32.81 ) that is a higher antioxidant ability than vitamin C antioxidant activity (RC50=51.21 ). DRE protected the hepatocytes against Fe3+/8-Hydroxyquinoline- induced cytotoxicity measured by trypan blue exclusion method. DRE also decreased ROS formation induced by Fe3+/8-HQ in hepatosytes as measurd by 2,7-dichlorofluorescein diacetate fluorescence. Also, mitochondrial membrane potential was restored in isolated hepatosytes after disturbance by Fe3+- HQ as measured by Rhodamine 123 fluorescence.
