بررسی چند شکلی های ناحیه پروموتر ژنIL7RAدر بیماران ایرانی مبتلا به اسکلروزیس چند گانه

Abstract

Multiple Sclerosis، یک اختلال مبهم اتوایمیون نسبتا شایع سیستم عصبی مرکزی میان افراد جوان است که بیشتر در جنس مونث دیده شده است که به علت پاسخهای التهابی منجر به دمیلینه شدن و تخریب اعصاب می گردد . این بیماری شایعمناطق معتدل است. IL-7RA یک نقش حیاتی در تکامل T-CELL های تیموسی و بقاء سلولهای خاطره محیطی بازی کرده است در نتیجه بعضی مطالعات MS را با تغییرات چند شکلی های تک نوکلئوتیدی در پروموتر ژن IL-7RA مرتبط می دانند. در این تحقیق سعی شده چندشکلیهای شایع در پروموتر ژن گیرنده اینترلوکین 7 مورد کاوش و بررسی قرار گیرد.روش ها: در این پژوهش ابتدا خون گیری و بدنبال آن استخراج DNA از گروه های بیمار(100 نفر) و کنترل سالم (80 نفر) صورت گرفت، سپس استخراج DNA و توالی یابینمونه ها انجام شده در نهایت اطلاعات و نتایج بدست آمده توسط نرم افزارSPSS17 مورد بررسی قرار گرفتهبود.یافته ها: فراوانی آلل ها با احتمال خطر بالا برای هر3SNP rs11567685, rs11567686 و rs7718919 میان کنترل ها و بیماران MS با مطالعات قبلی مرتبط است. یک ارتباط قوی میان آلل (T)rs11567685 T/C)504-) در ناحیه پروموتری IL-7RA وMS با 042/0p= وجود دارد. فراوانی بالای آلل (G) rs7718919 (5/87%) با 02/0 p= و آلل A)( در rs11567686(72%) با 01/0 p= در کنترلهای سالم نسبت به بیماران وجود دارد و این داده ها از این موضوع حمایت می کنند که آلل ماژور (G) rs7718919و آلل ماژور(A) rs11567686ممکن است یک نقش محافظتی در بیماران MS بازی کنند., Multiple sclerosis is an ambiguous common autoimmune disorderof central nervous systemamong young adults and mostly in female genuse that due to inflammatoryresponses leads to demyelinating of neurons.The widespread of this disease is in moderate temperature. IL-7RA plays crucial roles in T cell development in the thymus and survival of memory T cells in the periphery in conclusion,In this study have been tried to survey and explore the common polymorphisms in promoter of interleukin 7 receptor.Methods: This investigation gathered 100 patients and 80 healthy controls were sampled and after DNA extracting and sequencing of samples finally the data were analysed by SPSS 17 .Results: The frequencies of the high risk alleles for each 3 SNPsrs11567985, rs11567686 and rs7718919 among healthy and MS subjects were relative to previous studies.a significant link was found between the (T) allele of rs11567685 and MS(-504T/C) in the promoter region of IL-7RA.The higher allelic frequency of the (G) allele rs7718919 (87/5%), with (p=0/02 ) and the (A) allele rs11567686 (72%) with (p=0/01) exists among healthy subjects rather than MS patients and these data support the notion that the major (G) allele rs7718919 and majoar (A) allele rs11567686 may play a protective role in MS patients.
