آنالیز همراهی چند شکلی هایSNP309 و Arg72Pro به ترتیب در ژنهایMdm2 و P53 در بیماران مبتلا به سرطان کولورکتال

Abstract

سرطان کولورکتال یکی از شایع ترین بدخیمی ها در سطح جوامع بوده که شیوع آن در حال افزایش می باشد. این سرطان در زمینه عوامل محیطی و استعداد های ژنتیکی ایجاد می شود. مطالعات بسیاری ارتباط بین پلی مورفیسم های شایع ژن های P53 و MDM2 و سرطان کولورکتال را مورد بررسی قرار داده اند. هدف: ما در مطالعه حاضر به بررسی ارتباط احتمالی پلی مورفیسم های SNP309 MDM2 و Arg72Pro P53 با سرطان کولورکتال در جمعیت شمالغرب ایران می پردازیم. مواد و روش کار : مطالعه مورد شاهدی حاضر که برروی 100 بیمار مبتلا به سرطان کولورکتال و 100 فرد سالم بعنوان گروه شاهد انجام گرفت. از تمامی افراد مورد مطالعه خون محیطی اخذ گردید و پس از استخراج DNA، ژنوتیپ افراد از نظر دو پلی مورفیسم مذکور به روش PCR (polymerase chain reaction) مورد بررسی قرار گرفت. یافته ها: بررسی ژنوتیپی افراد نشان داد فراوانی ژنوتیپ های مختلف در پلی مورفییسم Arg72Pro P53 در گروه شاهد و مورد تفاوت معنی داری نداشت؛ همچنین پلی مورفیسم SNP309 MDM2 از نظر ژنوتیپی در دو گروه تفاوت معنی داری از نظر آماری نداشت., Colorectal cancer (CRC) as one of common malignancies in societies, results from the interaction between environmental exposures and genetic predisposition factors. Common polymorphism in the P53 gene and MDM2 gene may be risk factor for CRC. Aims: We designed a case control study to investigate the association between Arg72Pro of P53 polymorphism and SNP309 of MDM2 polymorphism with colorectal cancer in Northwest population of Iran. Methods and Materials: In this case control study, peripheral blood samples of 100 sporadic colorectal cancer patients and 100 normal controls were collected. DNA was extracted and genotyped for two polymorphisms using polymerase chain reaction technique. Results: Genotype analysis showed that the frequency of various genotypes in the polymorphism of Arg72Pro of P53 gene in two groups of case and control has not any significant differences. Also the frequency of genotypes in the other polymorphism (SNP309 of MDM2 gene) has not any statistically meaning different in case group compared to control group.
