ارتباط پلی مورفیسم های ژنی(T/G 2905 و -964 A/G)IL-27 با دیابت نوع 1 در منطقه آذربایجان

Abstract
Introduction: Type 1 diabetes (T1D) is an autoimmune disease results from the self reaction CD4+ and CD8+T cells that characterized by destruction of insulin-producing pancreatic cells. This disease occurs as a result of genetic and environmental factors. Interleukin 27 (IL-27) is a newly discoveredcytokine of IL-12 family that induced differentiation of Th1 and has been linked to the activation of CD8+Tcells and promotes humoral responses. So, IL-27 has a potential role in autoimmune diseases such as type 1 diabetes. Objectives: the aim of this study was to investigate allele and genotype frequencies of IL-27 (p28) subunit -964 A/G and 2905 T/G polymorphisms in type 1 diabetes compared to healthy controls. Materials and methods: DNA was extracted from whole blood samples of 110 type 1 diabetes patients and 302sex, age, and ethnically matched healthy control groups. IL-27 -964 A/G and 2905 T/G region of the IL-27 p28 subunit were amplified during PCR and analyzed by restriction fragment length polymorphism (RFLP). Results: IL-27 2905 T/G was not polymorphic in the study populations. The significant differences were found in allele and genotype frequencies of IL-27 -964 A/G polymorphism between type 1 diabetes and controls (p=0.007 and p=0.028; respectively). This difference was the result of a higher incidence of the IL-27 AA genotype in type 1 diabetes compared to healthy control group. In addition significant differences were not found in IL-27 -964A/G polymorphisms and sex or onset age of type 1 diabetes., هدف: هدف این مطالعه بررسی فراوانی آللی و ژنوتیپی پلی مورفیسم های -964A/G و 2905T/G از IL-27p28 در بیماران دیابتی و مقایسه ی آن با افراد سالم است.روشکار و مواد: از خون محیطی 110 بیمار دیابت نوع یک و 302 نفر افراد سالم که از نظر جنس، سن و نژاد با گروه بیماران هماهنگ بودند، DNAاستخراج گردید. ژنوتیپ های نواحی ژنی -964A/G و 2905T/G از زیر واحد IL-27 p28با روش PCR تکثیر شده و ژنوتیپ آن ها با روش RFLP مشخص گردید.یافته ها: در جمعیت مورد مطالعه ناحیه ی IL-27 2905T/Gپلی مورفیک نبود. مقایسه ی فراوانی آللی و ژنوتیپی IL-27 -964A/G در بیماران دیابت نوع یک با گروه کنترل تفاوت معنی داری با مقادیر ارزشی ( به ترتیب p=0.007 و p=0.028 ) نشان داد که علت این اختلاف، افزایش تعداد ژنوتیپ AA در بیماران بود. هیچ ارتباط معنی داری بین IL-27 -964A/G و جنس و سن شروع بیماری مشاهده نشد
