Investigation anticancer effects of probiotics originated from native microbiota on human cancer cell line KB

Abstract
Squamous cell carcinoma is the most common cancer in oral mucosa and radical surgery, chemotherapy, and radio therapy are considered as three common treatment methods. Recently probiotics have been assessed as effective anti-proliferative and anti-cancer agents and thus in this study, Acetobacter syzygii, Entrococus durans and Lactobacillus acidophilus strain secretions were used for cytotoxicity assessments on KB oral cancer cell line and KDR epithelial normal cell line Method and Materials: The cytotoxicity assessments on cells was performed through 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay as well as qualitative (4',6-diamidino-2-phenylindole staining) and quantitative (Flow cytometry assessments using the BD Biosciences Annexin V-FITC Kit) evaluations of apoptosis. Results: A. syzygii secretions showed 75.7 % apoptosis against KB cancer cell lines similar to cisplatin while apoptosis only was developed in 9.36 % of KDR normal cells. E. durans secretions showed 72.2 % apoptosis against KB cancer cell lines similar to cisplatin while apoptosis only was developed in 4.56 % of KDR normal cells.The anti-prolifrative effects of Lactobacillus acidophilus (PTCC 1643) as a reference bacterium, was similar to A. syzygii and E. durans indicating beneficial effects of useful bacteria impacts on oral disease prevention. (89.39% apoptosis on KB cancer cell lines and 6.91% apoptosis on KDR normal cells). Conclusions: The anticancer bioactivity of A. syzygii, E. durans and L. acidophilus strain secretions depends on the induction of apoptosis in KB cancer cells. However, several investigations should be conducted to precisely determine the effective compounds to be used as anticancer therapeutics in the future.
