Evaluating the cutotoxicity of orthodontic fixed retainer adhesives

Abstract
Bisphenol-A, is one of the chemicals that are released from the bonding and causes sensitivity, mutagencity and estrogenesity in body. Although studies of the liberalization of bonding that used in brackets are acceptable(0.1ppm) but no search is done about bonding of orthodontics fixed retainer that has more volume and more exposure time to saliva. The purpose of this study is to evaluate the cell cytotoxicity of orthodontic bonding fixed Retainers. Methods: Human gingival fibroblasts were prepared in Dulbecco's Modified Eagle's Medium (DMEM) culture For group 1, specific retainer binding light cure resin bonding composite (Transbond LR, 3M Unitek) to form 5/0x 1x1 mm discs (n = 12) between two glass slabs are prepared. For Group 2, restorative composite resin (Herculite XRV, Kerr / Cybron, Orange CA, US) to form 0/5x1x1 mm discs (n = 12) between two glass slabs prepared And with be cured about 20 seconds by the Light mw650 per square centimeter wavelength nm468. Group 3, control group and this group does not use any substance. The next step for removing layers of samples with large quantities of water will wash polymerization. After the cultivation Two hundred microliters of each sample Dvplykh and well plate 96 - will be put and the cells with 1.5 ml % 10 Alamar blue (Biosource, Camarilo, CA) and was incubated for one hour. The fluorescence intensity of each well using a fluorescent plate reader (FLUOstar Galaxy, BMG Lab technologies) at the excitation wavelength nm 530 and the emission measure will nm580. Possible differences between the four groups in terms of cell viability One-way ANOVA test or will be examined kruskal-wallis. If there are significant, appropriate post hoc test or U-Mann-withney test will be used (the first type of error is acceptable in this study, 0/05 is assumed.) results: The results show that, for viability in both Transbond LR and Herculite XRV significant differences with controls are (respectively, P <0.001 and P <0.001) but between Transbond LR and Herculite XRV significant difference in terms of cell survival was observed ( P = 0/105 ). conclusion: Transbond LR and Herculite XRV in this volume have similar cell cytotoxicity.
