Enterococcus faecalis is the pathogen usually found in endodontic infections and elimination of Enterococcus faecalis increases the success of treatment. Clinicians have been seeking speed and ease of preparation of root canal system for long. Therefore the object of the present study is comparing the single Rotary file (reciproc) and hand instrumentation in reduction of Enterococcus faecalis from the root canal of human extracted teeth. Material and methods: 72 single-human extracted teeth were divided in two equal experimental group and one control group (single Rotary file instrumentation (Reciproc) n=33 and the control group n=6). Two extra control groups (positive n=5, negative n=5) were selected for confirmation the canal infection, as well. All of samples were prepared by k-file #20 before sterilization. Then the teeth were autoclaved and infected with Enterococcus faecalis. The experimental groups were instrumented either with one file (Reciproc #40 6%) or hand instrumentation (Master apical file #40 and flaring #45-60). Bacteriological samples were taken after instrumentation to determine the level of remaining of bacteria. Result: 33 samples and 13 samples in hand file group and single file group, represented complete reduction of the bacteria respectively .There was a statistically significant difference between the two groups. Conclusion : Despite decreased preparation time with one file rotary, the bacterial debridement is not effective and conventional instrumentation is more efficient in elimination of Enterococcus faecalis from the root canal system. However, further clinical investigations are suggested in order to approve these results.
