Comparison of Antibacterial effect of photodynamic therapy and irrigation with sodium hypochlorite 2.5% using Real time-PCR technique in E.faecalis infected root canals
Abstract
The bacteria which enter the root canals of the teeth,invade the dentinal tubules and may cause viable infections in canals due to poor penetration of disinfectants into these tubules.Also bacteria like Entrococcus Faecalis can produce biofilms inside or outside the root canals which are difficult to eliminate.We can get access to complicated root canal systems by laser therapy,due to their adjustable penetration ability.Therefor the goal of this study was to compare the capability of photo activated laser and sodium hypochlorite (NaOCl)2.5 % irrigation in elimination of E.faecalis from the root canals by Real time PCR technique. Materials & Method: 60 extracted human central upper incisors were selected and sterilized by autoclve.Root canals were infected by E.faecalis and then incubated for 24 hours. Samples were devided randomly into 3 equal groups.Nothing was done on control group.In second group laser therapy with the power of 100 mW was performed by diod laser for 120 seconds.In third group canals were irrigated by 5 ml of Naocl 2.5 %,then all the samples were located in 15 mL of normal saline in test tubes and were transfered to sonicator for separation of the bacteria.On these samples DNA extraction was performed and then Real-time PCR technique was done. Results: According to the findings of this study,inhibition of bacterial growth in all experimental samples were significantly more than control group.There was a significant difference between photodynamic therapy and NaoCl 2.5 % statically. The effect of NaoCl in all samples was better than photodynamic therapy. The results of the average CT were 40 , 30.2 and 15.35 for 2.5 % Naocl , photodynamic therapy and control group , respectively. Conclusions: Based on the findings of this experimental study, NaoCl 2.5% eliminates E.faecalis in infected root canals effectively and this effect was more than that of photodynamic therapy
