Evaluation of changes in OIP5-AS1 expression and its relationship with miR-27 expression in laryngeal squamous cell carcinoma (LSCC)

Abstract
Cancer is a genetic disease in body cells due to abnormal cell division or loss of natural cell death (apoptosis). Today, cancer is known as the second cause of death after cardiovascular diseases. Laryngeal squamous cell cancer (LSCC) is known as the most common type of common malignancy in the larynx. Various environmental factors such as smoking, along with genetic factors, are known risk factors for this disease. Doing projects such as Transcriptome showed that most of the human genome is transcribed into non-coding RNA, and that these RNAs have different types that mostly have a regulatory role. These genes play different roles in regulating gene expression and various cell activities, and also play an important role in the growth and development of human cancers, including laryngeal cancers. Among these, the role of two non-coding genes OIP5-AS1 and its relationship with the expression level of miR-27 in different types of cancers have been studied and taken into consideration. Materials and methods: In this research, 50 samples of tumor tissue and 50 samples of tumor margins of cancer patients, including 45 men and 5 women, were examined. RNA was isolated from the target tissues using an extraction kit (trizol), cDNA synthesis was performed from the extracted RNA, then Real Time PCR was performed to express the target genes, and finally statistical analysis was performed by the software. Results: According to the results of the study, the expression of lncRNA OIP5-AS1 in tumor tissue was marginally increased compared to healthy tissue, and the expression of miR-27 was decreased. On the other hand, two were related to the clinical indicators of the gene, such as metastasis and lymph node involvement. Also, the findings related to the biomarker analysis showed that both genes have the separation of healthy tissue from the patient and have biomarker value.
