Recent studies have shown that chronic systemic inflammation can change the biogenesis pathway of exosomes. Considering the inflammatory nature of obesity, in this study, the biogenesis of exosomes in the liver tissue of male rats receiving a high-fat diet was investigated. Methods: 16 male rats were randomly divided into two groups including the control group (C) and the high-fat diet (HFD) group. In the high-fat diet group, the animals received a high-fat diet for three months. At the end of the third month, the animals were killed by injecting a high dose of ketamine and xylazine, and their liver tissue was removed for necessary investigations. Real Time-PCR was used to measure the expression of desired genes. Hematoxylin-eosin staining was used for pathological investigations. CD81 protein level was measured using western blot method. Results: Our results showed an increase in the expression of the inflammatory gene IL-1β, occurrence of pathological changes and an increase in the expression of genes related to the biogenesis of exosomes, including in the liver tissue of the high-fat diet group compared to the control group. Also, the level of CD81 protein in the liver tissue of the high-fat diet group was higher than that of the control group.en_US
