Targeting tumor-promoting factors in the tumor microenvironment has been one of the popular methods of immunotherapy in the last decade. There are many factors in the tumor area that help tumor growth and can even cause drug resistance and suppression of immune responses. Therefore, targeting these factors can be considered a suitable treatment solution. The il-6 axis and its receptor, gp130, is one of the very important reinforcement arms that has a noticeable increase in expression in cancer cells and contributes significantly to tumor growth. Therefore, in this study, it was decided to simultaneously suppress the expression of IL-6 and gp130 factors in cancer cells. Materials and methods: In this study, in order to inhibit the expression of IL-6 and gp130 factors, siRNA molecules loaded in chitosan-based nanoparticles were used. This study was conducted on two mouse cell lines, 4T1 and CT26. Examining the expression of target genes with Real-time method and checking cell viability with MTT method. Also, the ELISA method was used to check subtokine IL-6. Findings: The treatment strategy used in this study was able to reduce the survival of cancer cells and affected the expression of genes involved in survival. Also, the level of IL-6 cytokine secretion was significantly reduced.en_US
