dc.description.abstractBackground: Temperature-sensitive hydrogels have gained popularity due to their unique properties, including high water absorption, flexibility, and the ability to alter shape in response to temperature changes. With the increasing use of mesenchymal stem cells across various sectors, it is crucial to identify an appropriate culture medium for their growth and proliferation. By improving these hydrogels, we hope to enhance their suitability as a culture medium, thus supporting the effective growth and proliferation of mesenchymal stem cells for various biomedical applications.Purpose: This study aims to find a better scaffold by enriching beta-glycerophosphate chitosan collagen hydrogels. Methods: We create chitosan-beta-glycerophosphate collagen hydrogel with prepared platelet lysate and then characterise it using SEM and water absorbance tests. In the next phase, we combine the cells inside the synthesised hydrogel and set them in an incubator with 5% carbon dioxide gas for three and seven days to grow, after which we extract and segregate the cells for measurement. We do the MTT and viability tests.Results: The collagen-chitosan hydrogel enriched with platelet lysis provided consistent cell spacing, was morphologically appropriate, and did not differ from the other groups. Furthermore, the viability research revealed that the vitality of mesenchymal stem cells in the hydrogel was much higher than in other groups.Conclusion: The present work shows that the collagen-chitosan hydrogel, when supplemented with platelet lysate, enhances cell viability and possesses the capacity to accept cells within its structural cavities. This combination improves the hydrogel's suitability as a scaffold for cell culture, providing a more supportive environment for cell growth and proliferation.en_US
