dc.description.abstractIntroduction: Sunitinib is an oral inhibitor of tyrosine kinase. This is an effective therapeutic agent in the treatment of kidney cancer and gastrointestinal tumors resistant to imatinib. Determination of the concentrations of this drug in biological samples is very important for pharmacodynamic and pharmacokinetic studies and treatment evaluation.Purpose: The purpose of this study was to develop a simple, sensitive, and effective method for the determination of sunitinib in biological samples using high performance liquid chromatography (HPLC). Firstly, NiFe2O4/ MIL-101(Fe) metal-organic framework nanoparticles were synthesized by hydrothermal method, and it was used as the sorbent in extraction of the analyte from biological and water samples.Methods: For this purpose, a proper amount of the sorbent was added into the sample solution and the mixture was vortexed for 1 min. The sorbent particles were collected by the an external magent and the supernatnt was removed. To desorb the desired analyte, the sorbent was eluted with 100 μL DMF under vortexing. Then, the eluent was injected into the HPLC-MS/MS for quantitative analysis. Results: The method provided satisfactory results, including acceptable extraction recovery (76%), low limits of detection and quantification, accepatble relative standard deviations (≤ 6.9%), and a wide linear range. Finally, the proposed method was utilized as a sensitive method on biological samples including plasma and urine samples.Conclusions:This study has completely fulfilled its purpose; therefore, the proposed method can be successfully applied as a simple, sensitive, and cost-effective method in routine analysis for the determination of sunitinib in plasma and urine samples.en_US
