dc.description.abstract | Considering the increasing incidence of cancer in different societies, the design and implementation of new methods of cancer treatment is one of the most important research fields around the world. Immunotherapy based on targeting important factors involved in the growth and spread of cancer is one of the most obvious efforts of researchers in this field. CD73 and CXCR4 are the main factors involved in tumor growth. Combined inhibition of these factors in cancer can be considered an effective method in cancer treatment. As a result, in this study, it was decided to suppress the expression of these two factors in cancer cells using nanocarriers loaded with siRNA.
Materials and methods: In this research study, inhibition of CD73 and CXCR4 factors was suppressed in two mouse cancer cell lines including 4T1 and CT26 using nanocarriers loaded with siRNA. The effect of reducing the expression of these factors on cell viability was investigated by MTT method. Also, the effect of combined dermalin on the expression of target factors was evaluated by Real-time PCR method.
Results: Biodegradable nanoparticles based on chitosan were able to effectively deliver siRNA molecules to cancer cells and suppress the expression of CD73 and CXCR4 factors in both cell lines. This decrease in expression was associated with the induction of cell death and the decrease in the expression of factors effective in the growth and spread of cancer. | en_US |
