dc.description.abstractThe monitoring of therapeutic drugs involves measuring drug concentration in biological fluids. For this purpose, use of cheap, reliable, and fast analytical method is considered. Ofloxacin is a pyridine-carboxilic acid derivative of nalidixic acid, and is prescribed to treat infectious diseases such as urinary tract infections, prostatitis, pneumonia. The most common complications observed in the digestive system include diarrhea, vomiting, nausea. The common central nervous system complications include: headache, insomnia, and dizziness, According to these cases, it is necessary to control the concentration of ofloxacin in biological fluids such as plasma to prevent allergic effects or adverse toxicity in the patient.Aim:Extraction of ofloxacin from biological fluids, plasma, and measurement of ofloxacin in biological fluids by the electromembrane extraction-capillary electrophoresis (EME-CE) system.Method:To measure ofloxacin concentration in plasma by CE, ofloxacin must first be extracted and preconcentrated by EME. Considering that the basis of separation and extraction by EME is based on the electric field; Charged particles under the electric field are transferred from the aqueous medium like plasma sample to the membrane filled with organic solvent and then transferred to an aqueous acceptor phase and finally injected into the capillary electrophoresis.Results :In this work, the experimental conditions such as pH of donor and acceptor solutions, applied voltage, extraction time, stirring rate, organic solvent conditions were checked and optimized. The calibration curve was linear in the concentration range of 0.20 to 7.0 μg.mL-1 with a detection limit of 0.05 μg.mL-1 for plasma. The correlation coefficient of the curves was 0.9993. The accuracy of the method according to relative error is less than 8.65%.Conclusion:A simple, effective and reliable method for extracting and measuring ofloxacin in biological samples with appropriate accuracy and precision was presented.en_US
