dc.description.abstractUnfortunately, infertility is a very common medical problem. One of the side effects of chemotherapy drugs can be premature ovarian failure and finally infertility in these patients. One of the most widely used drugs in the field of chemotherapy is cyclophosphamide, whose adverse effects on the reproductive system of women have been investigated in research. Assisted reproductive techniques such as in vitro fertilization and freezing of ovaries or oocytes and maturation and development of ovarian follicles are among the most important strategies for maintaining fertility in women who have used the drug cyclophosphamide. Considering the possible mechanisms of the effects of this drug such as conditions of oxidative stress, use Antioxidants such as vitamin E can improve the success rate of maturation and follicular development in assisted reproductive techniques in patients who have received cyclophosphamide. Therefore, in this study, the effect of vitamin E added in the culture medium of antral follicles of mice treated with cyclophosphamide on oocyte maturation, fertilization and embryo formation was investigated. Method and materials: Adult female mice aged 6 to 8 weeks were divided into two main control groups and the cyclophosphamide treatment group, which received a dose of 150 mg/kg intraperitoneally. On the fifth day of treatment, they received 10 international units of Hmg to obtain antral follicles. And all the mice were euthanized on the seventh day after receiving cyclophosphamide to isolate and culture antral follicles by cervical vertebrae displacement method. Follicles were cultured in the culture medium containing and lacking vitamin E for 4 days, and on the fourth day, the level of BMP-15 and GDF-9 gene expression was analyzed by hemolysis of the follicles. In addition, by adding HCG in the culture medium of follicles, After 48 hours, the ovulation rate of GV, MI, MII oocytes was evaluated. Results and Conclusions: According to the results, it can be said that the maturation rate of mouse ovarian follicles, the percentage of fertilization and the percentage of two-celled embryos obtained from the antral follicles obtained from the ovaries increased in the culture medium containing 100 μM vitamin E. These results showed that vitamin E by reducing the level of free radicals, which is probably due to the administration of cyclophosphamide to mice and increasing the expression of maturation-related factors such as GDF-9 and BMP15 in cultured antral follicles, which ultimately increases the numbers MII oocytes and as a result the increase of two-celled embryos resulted from fertilization and in fact caused the survival of fertility potentially and this finding can be useful in the field of supporting reproductive techniques.en_US
