dc.description.abstractToday, osteoarthritis is a concern of many elderly people and professional athletes, and until today no definitive treatment has been introduced for this disease, and only drugs to control pain and inflammation have been marketed. In this study, we intend to use the anti-inflammatory properties of postbiotics for the purpose of controlling inflammation due to their unique characteristics. Materials and methods: First, K4IM cell line was cultured in DMEM culture medium containing 1% penicillin/streptomycin and 10% FBS, under sterile conditions under 37 degrees temperature and 5% CO2. After 24 hours, 10 ng/ml TNF-α was added. Unstimulated cells were also used as control group. Postbiotics were prepared with a standard protocol. Then these cells were treated with different concentrations of postbiotics. The optimal concentration obtained from postbiotics was added to them. After 24 hours, the cells were collected and their total RNA was extracted and cDNA was synthesized by the RNA extraction kit. The level of mRNA expression of the target genes was checked by qRT-PCR method and with specific primers, along with the internal control of beta-actin. Then, data distribution was checked using prism software and Test-T test was used for normal distribution and Whitney-Mann test was used for data with non-normal distribution. Results: In the cell line related to osteoarthritis, after crossing the cells with postbiotics extracted from yeasts, we had a significant decrease in inflammatory markers such as IL-1B, IL-6, IL-8, and CXCL-8, which results in gene expression levels were aligned. While anti-inflammatory markers such as IL-10, IL-4 and VEGF had a significant increase in gene expression levels after the effect of postbiotics.en_US
