Melanoma is one of the most common cancers with a high rate of spread, especially in areas with high intensity of light radiation, which also has a high mortality rate due to high metastasis. This cancer, along with pancreatic cancer, are known as the most lethal cancers with unclear pathogenesis, and efforts are ongoing to find the molecular mechanism and appropriate screening methods for patients with different prognoses. In this regard, in this study, we decided to investigate the changes in PTBP3, LncRNA BCRT1 and miR-1033 gene pathways in tumor samples and its possible changes in cancer samples. Methods: During this investigation, 50 tumor samples and 50 samples from the margin tissue around the tumor were collected as control samples. After collecting the RNA of the samples, it was extracted and the expression level of the desired gene was evaluated using the Real-time PCR method. The difference in the expression level of the two groups was investigated with appropriate statistical tests. Pvalue<0.05 was considered significant in statistical tests. Results: The results obtained from Real-time PCR showed that the expression of PTBP3 and LncRNA BCRT1 gene was significantly increased in tumor samples, while the expression of miR-1033 was significantly decreased in tumor samples. Regarding the pathological indices of all three investigated genes, they are related to lymph node involvement and the expression of PTBP3 and LncRNA BCRT1 genes with metastasis. Regarding biomarker value, only PTBP3 and miR-1033 genes have biomarker value in differentiating normal tissue from tumor.en_US
