dc.description.abstractFinding new ways to treat cancer is one of the most important goals of cancer researchers around the world. Cancer cells have diverse characteristics, including the ability to suppress antitumor immune responses and resistance to apoptosis. Therefore, monotherapy does not have the ability to control these diverse characteristics of cancer cells, and in treatment strategies, different characteristics of cancer cells should be targeted. CD73 molecule is increased on cancer cells and causes proliferation, angiogenesis, metastasis and suppression of immune cells. On the other hand, BV6 drug can induce apoptosis in cancer cells by suppressing apoptosis inhibitors. Therefore, the treatment of cancer cells by inhibiting the expression of CD73 and BV6 drug can be considered an effective method in cancer treatment. Materials and methods: This study was conducted on cancer cell lines including CT26 and 4T1. Biodegradable nanoparticles based on chitosan were used to transfer anti-CD73 siRNA molecules and BV6 drug to cancer cells. The effect of treatment on the survival of target cells was measured using MTT and Annexin V/PI methods. Also, the effect of treatment on the expression of target genes was studied using Real-time PCR method. Results: The results of the experiments showed that delivery of anti-CD73 siRNA molecules along with BV6 drug to cancer cells induced cell death in cancer cells. Also, combined treatment inhibited cell migration. In addition, combined treatment decreased the expression of genes involved in cell survival and increased the expression of apoptosis-promoting genes.en_US
