Exosomes are nanoscale extracellular vesicleswith a diameter of ~30 to 120 nmreleased into various body fluids by fusion of multivesicular bodies with the plasma membrane. Recent studies have revealed that chronicinflammation can alter the biogenesis of exosomes. Considering the inflammatory entity of overweight and obesity, this study was conducted toevaluate the biogenesis of exosomes in lung tissue of high-fat diet (HFD) male rats. Methods: In this study, 16 adults male Wistar rats weighing 190 ±10 g were randomly divided into two groups; Control group (C) and high-fat-diet (HFD) (each in 8). In HFD group, animals were received HFD for 3 months. Finally, animals were euthanized and exosme biogenesis were evaluated by measuring the acetylcholinesterase activity in BAL, expression levles of Rab27-b and Alix as well as CD63 protein levels in lung tissue. Results: Our finding showed a significant increase in the expression levels of Rab27-b and Alix and protein levels of CD63 in lung tissue of HFD group compared to the C group. Furthermore, data demonstrated that activity of acetylcholinesterase were increased significantly in BAL of HFD group compared to the C group.en_US
