Introduction: Rapid and functional replacement of lost or damaged tissues of the mucosa and mouth is one of the most important concerns of researchers and clinicians in the field of medicine and dentistry. The use of engineered tissue of the oral mucosa can be a suitable solution for treating and eliminating the defects or damages of the oral mucosa. During this study, the synthesis and design of adhesive mucus polymer scaffolds containing triamcinolone was carried out with the aim of investigating its anti-inflammatory and antioxidant effects. Methods and materials: Fibroblast cells were purchased from the National Center for Genetic and Biological Resources of Iran and cultured in three 6 cell plates. The scaffold was prepared using ethyl cellulose/gliadin polymer solution at room temperature. After disinfecting the scaffolds, a scaffold was placed inside each house. Then the cells were inflamed using lipopolysaccharide (LPS) of Escherichia coli (E.coli). After 48 hours, the amount of TNFα in the groups was titrated as an anti-inflammatory marker and the amount of DCF as an indicator of ROS in the presence of scaffolds and without scaffolds. Findings: The amount of TNFα measured in picograms per milliliter in the group containing scaffold with triamcinolone 097 was significantly decreased compared to the group of cells infected with LPS. The amount of DCF, which was an indicator of ROS (oxygen free radicals) in this study, was 3.90 mg in the group containing scaffold with triamcinolone, which was significantly reduced compared to the group of cells infected with LPS. Conclusion: In this study, ethyl cellulose/gliadin scaffolds containing triamcinolone have anti-inflammatory and antioxidant effects. Therefore, ethyl cellulose/gliadin scaffolds containing triamcinolone, due to their anti-inflammatory and antioxidant properties, can be used as a carrier for the drug triamcinolone in the treatment of oral inflammatory lesions.en_US
