dc.description.abstractColon cancer is a disease characterized by the uncontrolled growth of cells in the tissues of the colon. If left untreated, cell growth can spread outside the colon in a process called metastasis to the surrounding tissues or other parts of the body. Curcumin is a polyphenolic substance in turmeric rhizome that is a curcuminoid. Its antioxidant, antibacterial, anti-inflammatory and malignant inhibitory properties are used in the treatment of various diseases. Therefore, the aim of this study was to investigate the inhibitory effect of curcumin on MMP-9 gene methylation and migration of SW-480 cells in colon cancer. Methods: Colon cancer cells (SW-480) were cultured and treated with different concentrations of curcumin. Tetrazolium microculture (MTT) test was used to evaluate the anti-proliferative effects of curcumin on colon cancer cells. The migration of curcumin-treated colon cancer cells was also examined using the Scratch method. After extraction of whole DNA from treated and control cancer cells, the methylation of RECK gene promoter was investigated using MET-HRM method. Finally, the relative expression of different genes involved in cancer development and progression was evaluated using Real-Time PCR. Data obtained from real-time PCR were compared in control and treatment groups and the differences in expression of the studied genes were identified. Results : Findings from MTT test showed that the cytotoxic effect of curcumin was dose and time dependent. According to the results of this study, curcumin inhibits the migration of colon cancer cells by suppressing the 9-MMP gene. It was also observed that curcumin increases the methylation of the MMP-9 gene promoter in cancer cells. According to the results of the expression of genes involved in cancer, curcumin inhibits the growth and proliferation of colon cancer cells through various molecular mechanisms such as induction of apoptosis.en_US
