dc.description.abstract | Various factors are effective in increasing or decreasing resistance to chemotherapy in patients with Hodgkin's lymphoma. Therefore, recognizing these cases should be examined based on different populations. Accordingly, according to the different results obtained, the study examined the phenotype and genotype of patients with refractory Hodgkin's lymphoma and compared it with Hodgkin's lymphomas with response to treatment.
Methods: In this descriptive-analytical study, 80 adult patients with Hodgkin's lymphoma (40 responding and 40 refractory) were selected by convenience sampling. Samples were collected from all subjects in paraffin blocks. Finally, by adding the corresponding antibodies (CD117, CD30, CD15, P53, Ki67), the results were positive and negative. DNA of the sample was extracted by PCR, using primers pre-designed for bcl2, bcl6. The specific thermal cycler program was subjected to PCR and amplification, and finally the PCR product was electrophoresed on 3% agarose gel and was considered a positive sample if bands of a certain size were formed. Statistical analysis was performed by SPSS version 19.
Results: The mean age was 33.13± 15.29 in the response group and 35.95± 13.75 in the resistant group. CD117 in all patients was 80 (100%) negative, and CD30 was 80 (100%) positive. CD15 patients in the response group included 3 (7.50%) negative and 37 (92.5%) positive and in the resistant group included 28 (70.0%) negative and 12 (30.0%) P53, patients in the response group included 37 (92.5%) negative and 3 (7.5%) positive and in the resistant group included 15 (37.5%) negative and 25 (62.5%) was positive. The mean Ki67 was 47.75±17.90 in the response group and 70.50±16.48 in the resistant group. In the study of Bcl2 and Bcl6 genotypes, in all patients, 80 (100%) were negative. | en_US |
