dc.description.abstract | IntroductionIn the process of neuropathy induced by anticancer drugs, several mechanisms have been mentioned such as activation of the oxidative stress system, increasing the activity of N-methyl-D-aspartate(NMDA) receptors, and serotonin receptors.
Goal: The aim of the current study was to evaluate the preventive effects of Amantadine and Vitamin C on Cisplatin-induced neuropathic pain that was the result of cisplatin in the (spurious) mice.
Materials and methods:
Seventy-two male mice in a weight range of 25 to 35 g were randomly divided into 9 equal groups and underwent Amantadine and Vitamin C for 25 days. Different doses of Amantadine and Vitamin C were injected three days before injection of Cisplatin (5 mg/kg,IP ) at 4th day. Then injection of Amantadine and Vitamin C continued for 3 days. After that we have 3 days for rest. Finally we performed hot plate test. This 7 day cycle repeated 3 times for all groups. At day 25, blood samples were collected for Malondialdehyde )MDA( and و Total Antioxidant Capacity (TAC) Test.
Result: The results was showed a meaningful (p-value<0.05) increase in reaction to the pain after injection of these doses (20 mg/kg, IP) of amantadine and Vitamin C (100 mg/kg, IP) and also was observed the meaningful impact of Amantadin
( 5, 10, 20 mg/kg, IP) and Vitamin C( 100 mg/kg, IP) in these doses can decrease MDA level. Also, co-injection of the minimum doses of this drug had a statistically better effect than injection of each of them alone.
Conclusion: It is probable that a part of the inhibitory effects of Amantadine and Vitamin C is mediated by the decreasing of Inflamation in CNS. | en_US |
