Introduction: Valeriana officinalis (Valerian) is an ancient medicinal plant used for its various pharmacological influences such as sedative and analgesic effects on nervous system. Application of nanoparticles for stimulating production of secondary metabolites in plant invitro culture is a challenging issue. Iron is an essential substance for plant growth and metabolism. Therefore, it could be used as an efficient elicitor to affect production of secondary metabolites in in vitro plant culture. Aim: Investigating influence of modified nanoparticles of iron oxide with glutamic acid on phenolic and volatile compounds of Valeriana officinalis through invitro culture. Materials and Methods: The synthesized Glu-Fe3O4 NPs were characterized by using SEM-edx,TEM , DLS, FT-IR and XRD. In the invitro micropropagation of Valerian in the Murashige and Skoog (MS) medium, one month old Plantles were exposed to 5ppm, 10ppm and 15ppm of Fe3O4-Glu NPs; and after 30 days growth characteristics, total phenol and flavonoid content, photosynthetic pigments, antioxidant properties, phenolic and volatile compounds of the plant extracts were evaluated. Results: Elicitor treatments could induce production of some volatiles like elemene, endo-borneol and junipene in the n-hexane extracts of palntlets. . Treatment with 10 and 15 ppm of elicitor decreased number of leaves of seedlings and 5ppm treatment could increase number of stomatas in leaves in comparison to control. Conclusion: Our findings provided evidence that Fe3O4-Glu NPs in optimum levels may improve production of some volatile compouds and could be utilized as a novel elicitor for in vitro biosynthesis of precious metabolites in micropropagation of medicinal plants.en_US
