dc.description.abstractThe aim of this study was to evaluate the effect of serum-derived exosomes of patients with ankylosing spondylitis on T lymphocytes. Material and methods: Blood samples were taken from normal subjects and 1.077 mononuclear blood cells (PBMC) were isolated using a filament. Anti-CD11b, CD16, CD19, CD36 and CD56 antibodies were used to isolate T lymphocytes to remove dendritic cells, monocytes, granulocytes, B cells, platelets, erythroid progenitor cells and NK. Then, using the special kit to separate the exosome from the serum and according to the protocol, the exosome kit was isolated from the serum of patients with spondylitis encliosis. Measurement of cytokine expression of T lymphocyte subtypes before and after transfect with exosomes removed from serum of patients with spondylitis was evaluated using Real Time-PCR technique. Results: RT- PCR and ELISA data showed that the expression of Th17 (RORγt and STAT3) and Th1 (T-bet) transcription factors increased, respectively. Also, the concentration and expression of Th1 cytokines (IL-17, IL-23, TNF-α and INF-γ) were significantly increased in the group treated with exosomal T cells from patients with AS. . On the other hand, the expression of Treg transcription factor (FOXP3) and cytokines (TGF-β and IL-10) decreased.en_US
