Design and Fabrication of Niosomes Loaded Metformin for the study of Stem-ness Preservation of human Adipose Derived Mesenchymal Stem Cells

dc.description.abstract | Introduction Aging is the gradual decline in the tissues of organisms. Impaired quality and quantity of stem cells play a crucial role in aging, which could be alleviated by the intervention of metformin-loaded niosomes and preserve stem-ness capacity. Material Methods At first niosomes fabricated by thin film hydration technique and their size and morphology characterized by zeta-sizer and FE-SEM. After that cytotoxicity of metformin-loaded niosomes determined by MTT test and finally, effects of this delivery system on human-derived mesenchymal stem cells, determined through nanog, oct-4, rex-1, klf-4, sox-2, h-tert genes expression by rt-PCR technique at passage 7. Results: In this study, niosomes were fabricated using thin film hydration method with size of 175.9 nm, PDI: 0.149, zeta potential: -20.6, EE: 72%, Stability :72h in room temperature and Drug releasing: 65.81% through 72 h in 37 °C. No toxicity was reported with niosomes and metformin in clinical dose (100 µM). adipose-derived stem cells lost their stemness in passage 7 versus passage 3 by on average 19.9% Clinical dose of Metformin (100µM) improved stemness gene expression by 4.65% And nano-Niosomal loaded metformin enhanced this effect by 7.35%. Conclusion: Metformin-loaded niosomes have been shown that enhance the stem-ness preserving effect on Adipose-derived stem cells. | en_US |
