dc.description.abstractIn this study, cellular toxicity and side effects of methotrexate loaded Cat-MNPs on Saos-2 cell line and mouse model have been studied respectively. Materials and Methods: At first Ts-CD was synthesized and then alylimidazole was used for surface modification.. After polymerization, MTX was loaded on the nanocarrier.The designed nanoformulation were characterized and its drug loading capacity and drug release profile were studied as well nanocarrier’s biocompatibility was proved by hemolysis assay. Efficacy of the nano-formulation in killing cancer cells was assessed using MTT assay. Finally, the side effects of the drug –loaded nanocarrier and free drugs were assessed using the mouse model. Results: Our data showed that the Cat-MNPs exhibit appropriate physicochemical characteristics. Drug release data indicated higher rate of drug release in acidic environment (pH= 5.4). Hemolysis assay confirmed the Cat-MN is a hemo-compatible nanosystem. MTT assay showed that nanoparticles lacking drug have no significant toxicity while nanoparticles containing MTX caused higher toxicity. Moreover, in-vivo study showed no significant side effects.en_US
