Abstract: Background & aim: cycle breast pain refers to moderate to severe pain one or both breasts lasting over a five-day period and faded after menopause and leads to various complications in women’s lives. Evidence on the effect of vitexagnus and flaxseed on cyclical breast pin (mastalgia) is not enough. This study aimed to assess the efficacy of vitexagnus and flaxseed on cycle mastalgia. Methods & materials: this triple blind randomized controlled trial was conducted on 159 women referred to health centers of Tabriz during October 2013 until September 2014. Subjects were allocated into three groups (n = 53 per group) using block randomization. Group I received 25 grams daily flaxseed powder and placebo of vitexagnus; group II received daily3.2-4.8 mg vitrxagnus tablet and placebo of flaxseed and control group received both placebo. Nominal day breast pain (NDBP), pictorial blood lost Assessment chart (Hingham), the shortened premenstrual (PAF) were applied at baseline, 1, and month after the intervention. Data was analyzed using general linear model. Results: one subject dropped out in the flaxseed group at the end of the first month after the intervention. Mean breast pain before the intervention was equal to 12.44(SD: 4.7) among all subjects. The breast pain improved significantly in both intervention groups during the first and second month after intervention. Mean NDBP score was significantly lower than that in the control prop at the first month after the intervention in the flaxseed group [adjusted mean difference: -3.1(95%CI:-4.2to -2.0)] and vitexagnus [-3.3(-4.3 to -2.2)] and the second month after the intervention in flaxseed [-7.0(8.1 to -5.9)] and vitexagnus [-6.4 (-705 to -503)]. The menstrual bleeding reduced significantly in both intervention groups during the first and second month after intervention. Mean high am score was significantly lower than that in the control group at the first months after the intervention in the vitexagnus group [-7.9(-11.8 to -4.0] and flaxseed [-9.6(-1305 to -5.6] and the second month after the intervention in vitexagnus [-10.1(-15.20to -5.0] and Flaxseed [-10.4(-15.5 to -5.2]. there was no significant difference between vitexagnus and flaxseed groups. Also the premenstrual symptoms improved significantly in both groups during the first and the second month after the intervention the mean total PMS score was significantly lower than that in the control group at the first months after the intervention in the vitexagnus group [-3.3(-4.0 to -2.1)] and flaxseed [-403(-5.5 to[-3.3(-4.0 to -2.1)] and flaxseed [-403(-5.5 to -3.0] and the second month after the intervention in vitexagnus [-5.8(-7.0 to -4.7] and flaxseed [-6.6(-8.1 to -5.7] there was no significant difference between vitexagnus and flaxseed groups. Conclusion: flaxseed and vitexagnus are effective in short-term period in decreasing cyclical breast pain (massages), reduction of menstrual bleeding amount and improvement of premenstrual syndrome. However, further studies are needed to examine the long-term effectiveness, lasting effect and adverse effects of this medication after stopping taking medication in order to decide whether this medication is suitable to treat mastalgia, reduction of menstrual bleeding amount and improvement of premenstrual syndromes or not.en_US
