dc.description.abstractThe mouth is the entrance to the digestive tract, which plays an important role in the initial reception and digestion of food (1). Rapid and functional replacement of missing or damaged tissues of the oral mucosa is one of the most important concerns of researchers and clinicians in the field of medicine and dentistry(4). Failure to repair deep wounds, especially in the mouth, can eventually lead to further infection and suffering of the patient (15). The purpose of this research is to synthesis and evaluate the anti-inflammatory properties of scaffolds containing curcumin and chrysin to accelerate the healing process of oral mucosal ulcers. Materials and methods These scaffolds have been synthesized based on the previous defended thesis of the presenter and have passed the relevant physical and mechanical tests. An executable example of these scaffolding is available. Based on the defended thesis, MIC of two plant extracts was detected, curcumin and chrysin, and is located in the structure of polymeric decomposable scaffolds synthesized with FDAapproved materials. The polymeric materials of these scaffolds include gelatin, collagen and alginate. The same scaffolds have been used in two groups with and without curcumin and chrysin extracts in this study.OSCC cells with the code c11066 were purchased from the National Cellular Reserve Center of Tehran and placed on the scaffolding after disinfection. After 48 hours, The cell surface environment was collected and examined by ELISA abcam-ab181421 kit with ELISA test to determine the amount of TNFα in the control and control groups. The statistical data obtained were compared between the two groups by independent t-test or Mann-Whitney nonparametric test and SPSS software version 17. Results According to the results of this study, the TNFα level in the SCC cell culture medium was 24.22 pg/ml in normal conditions, 5.6 pg / ml in the case of curcumin scffolds and 8.25 pg / ml in the case of chrysin containing scaffolds. The amount of TNFα in the PCL / Gel / CUR group is significantly reduced compared to the control group and PCL / Gel.The amount of TNFα in the PCL / Gel group is significantly reduced compared to the control group. The amount of TNFα in the PCL / Gel / CH group is significantly reduced compared to the control group and PCL / Gel.There is no significant difference between TNFα levels in PCL / Gel / CH and PCL / Gel / CUR groups. Conclusion: According to the results of this study, scaffolds containing curcumin and chrysin extract with MIC = 1000ppp reduce the secretion of TNFα by SCC cells.Due to the significant effect of chrysin and curcumin on the rate of TNFα secretion, curcumin and chrysin can be used in oral ulcers.en_US
