dc.description.abstractIntroduction: Opioid dependence is one of the major problems in the chronic use of these drugs. In this context, finding a way to curb dependence or reduce symptoms of cessation of consumption will significantly help to prevent dependence and quit the addiction, especially in the detox phase. Aim: The aim of this study was to evaluate the effect of chronic administration of E. macrophylla extract on morphine withdrawal in male rats. Materials and Methods: The E. macrophylla methanolic rhizome extract was used to investigate its effects on morphine withdrawal syndrome in male rats. 56 male wistar rats weighing 225- 275 g in 6 groups (n= 8 in each group) were used including morphine control group, saline control group, groups receiving three different doses of E. Macrophylla extract (100, 200, 400 mg/kg) with morphine, and the last group received the most effective dose of the extract (400 mg/kg) without morphine. To induce dependency, increasing doses of s.c. morphine was injected for nine days. Locomotor activity test was done for two groups: the effective dose of extract + morphine and saline + morphine groups in the 9th day. One hour after morphine, naloxone was injected and the effect of the extract on withdrawal syndrome was evaluated. Also, the levels of MDA and TAC were evaluated with related procedures. Results: Methanolic extract of E. macrophylla (200 mg/kg and 400 mg/kg with p<0/01 and p<0/001 respectively) decreased total withdrawal syndrome of morphine as compared to the normal control group. The extract (400 mg/kg) + morphine significantly decreased MDA level as compared to the morphine + saline group. Conclusion: I.P. injection of methanolic extract of E. macrophylla was effective in decreasing morphine withdrawal syndrome in male rats.en_US
