| dc.description.abstract | squamous cell carcinoma (SCC) is one of the most prevalent malignancies of the oropharynx. Recently. Many studies have emphasized the role of genetic and epigenetic impairments in development of malignancies. The role of ITIH5 and FBLN1 genes has been established in variety of malignancies, however, few studies have investigated their role in SCC. Therefore, we aim to evaluate the role of methylation of these genes in SCCMethods & Materials:In this study which was conducted as a case-control study, specimens were extracted from tumor tissue and adjacent area of tumor (0.5 to 30 mg) of those patients with tongue SCC, for molecular evaluations. The number of specimens was 20 ones from tumor and adjacent area of it (overall 40 specimens). The evaluation of methylation was conducted with bisulfitization of DNA and thereafter, semi-quantitative evaluation by means of Bisulfite Direct Sequencing MS method. Then, analysis of the data was done by SPSS version 22. Results:An overall 40 specimens from 20 cases were evaluated who the majority of them (60%) aged over 55 years and frequency males were more than females (11 vs 9 patients). The Rate of methylation of FBLN1 in tumor tissue was 63.27% and in control specimens were 36.73%. Also, cells with methylation of FBLN1 were significantly more prevalent among tumor cells (p=0.0159, odds ratio=2.901). The rate of methylation of ITIH5 in tumor tissue was 30% and in control tissue was 15%. Also, the rate of cells with methylation of ITIH5 and without methylation were 30% vs 20% respectively (odds ratio= 3.5, p-value 0.0046) | en_US |
