dc.description.abstractNo comprehensive information about uterus fatty acids (FAs) and prostaglandins (PGs) signaling changes during implantation period is available. In this study we evaluated effects of seminal vesicle secretion and diet on them. Methods: In this study, 120 female mice were randomly divided into six groups: control, seminal vesicle excised (SVX)-mated, ω-3 and -6 fed, SVX-mated+ω3-fed and SVX-mated+ω6-fed. Phospholipid FAs composition (using gas chromatography), PGI2, PGD2 and 8-iso-PGF2α levels with ELISA and protein levels of their receptors (IP, DP and FP) by western blot, as well as the synthesizing enzymes expression by qRT-PCR were evaluated in uterus during the implantation window. Results: The levels of linoleic acid (LNA) and arachidonic acid (ARA) increased about 50% on the fourth and fifth days of pregnancy. Moreover, higher levels of the PGs synthesizing enzymes mRNA, the receptors protein and the PGs levels were observed in the implantation day compared to the pre-implantation days (p<0.05). Our results showed that mating with SVX male could decrease ARA levels (about 25% on day 5), expression of the genes, as well as the PGs levels especially PGI2 and PGF2α (about 23% and 25% on day 5, respectively) in the uterus and so could reduce the implantation rate. In contrast with ω3, ω6-enriched diet significantly increased the genes expression and the associated PGs levels (p<0.05).en_US
