dc.description.abstractAim: Type 2 diabetes is a complex disorder that is affected by genetics and environmental factors. The human gut flora affects human health by producing different materials. Pattern of gut microbiota is influenced by food intak. Inulin is a fructo- oligosacarid that influences on gut flora and induces a beneficial pattern of flora in intestine. Mir-375 is a nucleotide structure that has great impact of several proteins, improves pancreatic β cell function and shows the preqnession of diabetic complication. KLF5 is one of the target genes of Mir-375. Mir-375 influences on KLF5 transcription and suppresses the production of KLF5. KLF5 has an underling role in stimulation of inflammatory cascades and cardiovascular complication. According to with these pointes the aim of this study was to evaluate the effect of inulin supplementation on mir-375 concentrations in plasma and KLF5 expression in PBMCs of patients with type 2 diabetes. Method and Materials: Forty- six type 2 diabetes patients (19 male, 27 female) were enrolled in this study. All subjects were randomly divided into two placebo and intervention groups. The intervention group (n = 23) received 10 g inulin supplement twice daily for 6 weeks. The control group (n = 23) received a starch-based placebo to consume twice daily for 6 weeks. Anthropometric parameters, blood pressures, Demographic and dietary intake were evaluated before and after 6 week of intervention. Also 10 ml blood samples were collected in initiate and end of study. Then plasma and PBMCs were separated and kept at -70 c. plasma levels of mir-375 and expression of KLF5 in PBMCs were measured using RT-PCR Statistical analysis was performed with SPSS version 23. Results: Intragroup analysis showed that inulin supplementation significantly reduced Waist circumference, Hip circumference, fasting blood sugar and diastolic blood pressure (p<0.05). There were no significant differences in anthropometric measurements and biochemical concentration throughout the study (p>0.05). Between - groups analysis in this study showed inulin supplementation has a significant effect on fasting blood sugar. Inulin supplementation lead to increase in plasma mir-375 and decrease in expression of KLF5 in PBMCs in comparison with placebo group. Conclusion: inulin supplementation is effective on plasma concentration of mir-375 and KLF5 expression and these factors are considerable in improvement of glycemic status of diabetic patients.en_US
