dc.description.abstractSelegiline hydrochloride is a selective irreversible MAO-B inhibitor which is used in the treatment of early stage Parkinson's disease, depression and dementia. It shows low bioavailability due to a high hepatic first-pass metabolism. The present work was undertaken to formulate mucoadhesive buccal films of selegiline (Sel) to improve the related therapeutic efficacy, patient compliance and bioavailability. Mucoadhesive buccal films of Sel were prepared by a solvent casting technique using hydroxypropyl methylcellulose (HPMC), Eudragit RL 100, carbopol 934p (Cp) and polyvinyl pyrolidone. The prepared films were evaluated on various physicochemical parameters such a weight, thickness, surface pH, swelling index, residence time, folding endurance, in vitro release, permeation studies and drug content uniformity. The films exhibited a controlled release over more than 8 h. It was concluded that the films containing 50 mg Sel in a polymeric mixture composed of 23 % carbomer 934p and 77 % HPMC exhibited satisfactory swelling, optimum mucoadhesive strength and a promising drug release.
