بررسی پلی مورفیسم های ژن TP53 (Arg 72 Pro, intron3 duplication 16 bp)و پلی مورفیسم ژن XRCC3 (Thr241Met)در زنان مبتلا به سرطان پستان در استان آذربایجانشرقی ایران

Abstract

Objectives: The investigative aim of this study was to examine and define the role of two polymorphisms in p53 gene (changes in exon 4 and intron 3) and polymorphism in exon 7 of XRCC3 may be associated with an increased risk for breast cancer in the female patients from the Eastern Azerbijan of Iran. Methods: 100 women with breast cancer and 100 healthy women without any background of cancer were included in the presnet study. Genotyping was performed by PCR_RFLP (polymerase chain reaction-restriction fragment length polymorphism) method employing the Bstu and Nla restriction enzymes following to DNA extraction. The results was subjected to statistical analysis. Results: Statistical analysis of the results obtained in the present study did not show a significant correlation between polymorphisms of TP53, R72P (P =0.36) and PIN3, Ins16bp (P = 0.13) and breast cancer in the population of East Azerbijan of Iran. No significant association were also found between XRCC3 gene polymorphism and breast cancer (P = 0.42)., هدف:در این مطالعه تعیین فراوانی دو پلی مورفیسم در ژن)TP53اگزون 4 و اینترون 3)و پلی مورفیسم اگزون 7 ژن XRCC3در بروز سرطان پستان در جمعیت آذربایجانشرقی ایران مد نظر قرار گرفت.مواد و روش ها: تعداد 100 نفر بیمار مبتلا به سرطان پستان و 100 نفر از افراد به عنوان کنترل که سابقه هیچ نوع سرطانی را در خود و خانواده شان نداشتند در این مطالعه شرکت داده شدند. بعد از استخراج DNA از نمونه های بیمار و کنترل و انجام PCR، پلی مورفیسم ها ی مورد نظر به روش PCR-RFLPو با استفاده از آنزیم های Bstu وNla مورد بررسی قرار گرفتند و یافته ها با روش های آماری مورد انالیز قرار گرفتند.یافته ها: بررسی آماری نتایج حاصل از این مطالعه ارتباط معنی داری بین پلی مورفیسم های ژن TP53،R72P (36/0=p) و PIN3Ins16bp (13/0=p) و سرطان پستان در جمعیت آذربایجان شرقی نشان نداد، همینطور آنالیز آماری ارتباط معنی داری را بین پلی مورفیسم ژن XRCC3 و بروز سرطان پستان نمایان نساخت (42/0p=).
