حذف 4977 جفت بازی در DNA میتوکندریایی و ارتباط آن با تعداد نسخه های DNA میتوکندریایی در فرآیند پیری

Abstract

Background:as one of the most frequent somatic mutations accumulated during aging in human mitochondrial DNA, the 4977bp deletion has intrigued scientific interest in recent years. Although many studies have shown a significant increase in the amount of 4977bp deletion, the findings with respect to an age-dependent escalate of mtDNA4977 in blood are still disputatious. Material and Method: we investigated the presence of common deletion using Multiplex PCR in whole blood samples of 100 elder individuals (6090 years). For detecting the mtDNA content and the percentage of deletion used Real Time PCR methode. Result: We detected the accumulation of common deletion in 46 elder individuals. the mtDNA content in elder individuals with 4977 bp deletion decreased nearly 3 fold and there was statistically significant difference between the aged and young individuals in mitochondrial content (P=0.01) and deletion levels ranged from 2% to 17% of the total mtDNA (mean: 10%0.02%)., هدف: هدف از انجام این مطالعه بررسی وجود حذف 4977 جفت بازیدر افراد سالمند و اثر آن بر تعداد کپی های mtDNA بود.روش کار و مواد: نمونه خونی از 100 نفر سالمند جمع آوری شد و پس از استخراج DNA با انجام Multiplex PCR وجود حذف 4977 جفت بازی بررسی شد. سپس با استفاده از تکنیک Real Time PCRبررسی اثر این جهش بر روی تغییرات کپی های mtDNAو مشخص شدن درصد حذفصورت گرفت.یافته ها: تجمع 46%از این حذف در 100 فرد سالمند مورد بررسی دیده شد.بررسی ها کاهش تقریبا 3 مرتبه ای را در محتوای mtDNA سالمندان نسبت به جوانان نشان داد. از لحاظ آماری نیز تفاوت معنی داری در محتوای mtDNAبین سالمندان و گرو کنترل جوانان دیده شد. (p value 0.01) و درصد حذف شدگی 2%تا 17%(با میانگین 10%) محاسبه شد.
