Recently,a new endodontic cement(Calcium Enriced Mixture)has been introduced using MTA analoge ,and its antibacterial effect seems better than MTA. Moreover, chlorhexidin usage increases for promoting antibacterial effect of dental materials and improving prognosis. This study deals with experimental examination of the effect of adding chlorhexidin to CEM cement on its antibacterial effect. Materials and Methods: Antibacterial activity of under experiment material(CEM Cement +CEM Fluid) and (CEM Cement+CEM Fluid+chlorhexidin 2%)against E.Faecalis , S.aureus , P.aeruginosa and E.coli is evaluated using agar diffusion method. Then diameter of the zone of inhibition around experimental materials is calculated by three observers after 24,48 and 72 hours. Findings: In the present study, mean diameter of the zone of prohibition in groups(CEM Cement+CEM fluid) and (CEM Cement+CEM Fluid+chlorhexidin 2%) against P.aeruginosa, E.Faecalis, S.aureus, E.coli was obtained as(13/2,9),(21/10,6),(20/2,9),(17,9/75)mm, respectively which in all of these groups, diameter of zone of inhibition in group (CEM Cement+CEM fluid+chlorhexidin 2%) was significantly greater than group(CEM Cement+CEM fluid). Conclusion: Adding chlorhexidin to CEM fluid increases antibacterial effect of CEM Cement
