dc.description.abstractDialog of oral cancer suggests it is one of the most prevalent cancers in the world. A combination of chemotherapy and nanoparticle drug delivery have shown significant promise in cancer treatment. There are some challenges and design specifications that need to be considered in optimizing nanoparticle-based combination chemotherapy. Methods: In this study, 75 male rats were divided into five groups. Newly formulated doxorubicin-methotrexate-loaded nanoparticles and free doxorubicin were IV-administrated to rats with 4-nitroquinoline-1-oxide induced oral squamous cell carcinoma. Results: Results showed that DOX-MTX- nanoparticle complex caused significant (p<0.05) decreases in the severity of histo-pathological changes, cellular proliferation, and death rate when compared to free DOX. There was also better weight gain, though not significant, in the nano-complex group compared to the DOX treated group. Conclusion: DOX-MTX-nanoparticle complex is more effective than free DOX in chemotherapy treatment of OSCC in rat models. However, further investigations are necessary to clarify the advantages and disadvantages of the complexes and its potential therapeutic application in different types of cancer
