Abstract

Abstract Background: Nowadays, Drug delivery systems have derived many attentions due to the decreasing in the side effect of drug. Nano gels are a type of drug delivery system that has many applications. Chitosan is a biocompatible and biodegradable polymer which can form ionic interactions through its amine groups and can be utilized in preparation of TPP nanoparticles. Chitosan is a physical cross linker. Scope: In this study, Insp6 molecule has been used as an individual cross linker for preparation of chitosan. Characteristics including Size, zeta potential value, cytotoxic effect, cell uptake and drug loading of the MTX containing Nano gels that have been prepared by TPP and Insp6 were compared. Methods: Size and zeta potential of the particles were measured by DLS. Drug loading was investigated indirectly by UV-Vis spectroscopy. Cell uptake of the drug was studied by flowcytometery and cytotoxic effect was evaluated by MTT method. MCF-7 was the cell line used in this study. Results: Insp6 cross linked Particles were larger in size, zeta potential of these particles was -6.97, completely different from zeta potential value of TPP-cross-linked nanoparticles. Drug loading of the Insp6 cross-linked nanoparticles was more than the TPP cross-linked ones. Cell uptake of the two nanoparticles did not show any significant difference. Both nanoparticles did not cause any cytotoxicity. Conclusion: This study was done for introducing of Insp6 as a physical cross-linker for chitosan. Insp6 can be an appropriate cross-linker for chitosan nanoparticles. More studies can help to prepare particles with size and physical characteristics suitable for certain applications.
