Comparison of protective effects of dl-Dunnione and Tangeretin against cisplatin-induced Nephrotoxicity in male rats

Abstract

This investigation aimed to appraise potential protective effects of dunnione and tangeretin, against cisplatin-induced nephrotoxicity in male rats. Methods: 48 male wistar rats were recruited in this study. In respect with tangeretin, twenty-four rats were randomly divided into 4 groups; 1- healthy control group, 2- Cisplatin only, 3- cisplatin plus tangeretin and 4- tangeretin. Tangeretin was injected intraperitoneally 5 mg/kg dose for 10 days, and a single dose of cisplatin (8 mg/kg) was injected on the 7th day. In respect with dunnione, twenty-four rats were randomly divided into 4 groups; 1- healthy control group, 2- Cisplatin only, 3- cisplatin plus dunnione and 4- dunnione. Dunnione was administered orally at 20 mg/kg dose for 4 days, and a single dose of cisplatin (20 mg/kg) was injected intraperitoneally 12 h after the first dunnione administration. Inflammatory/oxidative stress/apoptotic markers, kidney function, and urinary markers of renal injury were assessed. Results: Tangeretin and dunnione repressed cisplatin-induced nephrotoxicity in kidneys as indicated by decreased urea, creatinine and NFkB patheay mediators as well as by increased Nrf2 pathway mediators. Dunnione administration promoted NAD+/NADH ratios followed by enhancing the activities of SIRT-1 and PPAR-1; meanwhile, attenuated nuclear acetylated NF-kB p65
